Pharmaceutical co‐crystal: An alternative strategy for enhanced physicochemical properties and drug synergy

A growing number of co-crystals in the literature are proof how significant co-crystallization concept has become. Co-crystallization enhances physicochemical properties through formation intermolecular interactions between a drug and co-former. co-crystal is single crystalline material consisting at least two molecular components solid room temperature present definite stoichiometric ratio. Pharmaceutical consist active pharmaceutical ingredient co-former selected from generally regarded as safe (GRAS) list United State Food Drug Administration. Co-crystal requires an understanding target, proper choice only achieved experimentally after several trials. Other beneficial outcomes include binary eutectics, dispersions, amorphous forms, etc. Several key issues including design strategies, selection, methods; tradition newly synthetic methods that more efficient suitable for large scale have been briefly described. The preference demonstrated with particular emphasis on multidrug their contribution to combination strategies used treatment management resistance adverse side effects serious medical conditions require administration high doses such HIV/AIDS, tuberculosis, others.
